Different aspects of the tumor microenvironment
are studied, including hypoxia, the end-products of the metabolism and the
recruitment of host-derived cells. The major questions addressed are how
these different parameters and/or actors influence tumor angiogenesis,
metastatic progression and tumor response to conventional anti-cancer
therapies (ie, radio-, chemo- and immunotherapy). Major current projects are: 1) Study of the role of intermittent vs. continuous hypoxia in tumors and in particular,
through transcriptomic and proteomic studies, their differential impact on
(i) the phenotype of tumor endothelial cells and the consecutive
resistance to anti-angiogenic treatments and (ii) the metastatic potential
of tumor cells and their resistance to apoptosis. 2) Study of the tumor metabolism end-products,
including (i) the role of lactate as a (recycled) energy fuel for
oxidative tumor cells and (ii) the reconversion of nitrite into NO
under tumor low pH and low pO2 conditions and the therapeutic
and pathogenic consequences. 3) Study of the role of hematopoietic and immune cells
(macrophages and lymphocytes) on the tumor progression, with a focus on
the vasculogenic pathways and the immunosuppressive mechanisms in tumors.

This research
is directly derived from the expertise acquired through basic research: 1) Study of
the serological and seric proteome
of tumor-bearing mice and cancer patients to identify circulating tumor
auto-antibodies or proteins as biomarkers of cancer progression and
response to treatments. 2)
Development of tumor
vascular-targeting nanoparticles to deliver therapeutic entities such
as radio-isotopes and siRNA to tumors. 3)
Implementation of a technological
platform to identify anti-angiogenic and anti-tumor compounds issued
from hypotheses-driven neosynthesis of chemicals or
ethnopharmacology-selected plant extracts.

completed with conventional and high-standard molecular biology,
biochemistry and cell biology equipments. A variety of mouse models of
human tumor xenografts and metastases are also available.
